چکیده
Macropinocytosis is a ubiquitous cellular uptake mechanism of peptide-based intracellular delivery. This entry pathway shows promise as route for the biomacromolecules and nanoparticles. In this work, we obtained 8-residue analogue P4A bearing higher macropinocytosis induction ability. contains vital cysteine residues in its sequence, which immediately reacts with cystine culture medium to convert into oxidized forms, including intramolecularly form (oxP4A) dominant active species. The conjugate oxP4A membrane lytic peptide LK15 delivered bioactive proteins cells; notably, functional fused negatively charged protein tag at significantly reduced amount (up nanomolar range) without compromising delivery efficiency activities proteins.

has been synthesized as a conformational model for disulfide loops of limited ring size. 'H-nmr studies a t 270 MHz establish the presence of three intramolecular hydrogen bonds involving the Leu, Val, and methylamide NH groups in CDCl,. Evidence for peptide aggregation in CDCl, is also presented. A structural transition involving loosening of the hydrogen bond formed by the Val NH group is obs...
متن کاملSynthesis of bicyclic organo-peptide hybrids via oxime/intein-mediated macrocyclization followed by disulfide bond formation.
A new strategy is described to generate bicyclic peptides that incorporate non-peptidic backbone elements starting from recombinant polypeptide precursors. These compounds are produced via a one-pot, two-step sequence, in which peptide macrocyclization by means of a bifunctional oxyamine/1,3-amino-thiol synthetic precursor is followed by intramolecular disulfide formation between the synthetic ...
متن کاملDiscovery and Characterization of a Disulfide-Locked C2-Symmetric Defensin Peptide
We report the discovery of HD5-CD, an unprecedented C2-symmetric β-barrel-like covalent dimer of the cysteine-rich host-defense peptide human defensin 5 (HD5). Dimerization results from intermonomer disulfide exchange between the canonical α-defensin Cys(II)-Cys(IV) (Cys(5)-Cys(20)) bonds located at the hydrophobic interface. This disulfide-locked dimeric assembly provides a new element of stru...
متن کاملThe interaction of thioredoxin with Txnip. Evidence for formation of a mixed disulfide by disulfide exchange.
The thioredoxin system plays an important role in maintaining a reducing environment in the cell. Recently, several thioredoxin binding partners have been identified and proposed to mediate aspects of redox signaling, but the significance of these interactions is unclear in part due to incomplete understanding of the mechanism for thioredoxin binding. Thioredoxin-interacting protein (Txnip) is ...
